江赐忠,男,1974年生,福建人,长聘教授,二级教授,博士生导师,国家重大科学研究计划首席科学家,同济大学生物信息系特聘教授,同济大学转化医学高等研究院教授,课题组组长。
南开大学学士与硕士,美国爱荷华州立大学(Iowa State University)遗传学博士。毕业后先后在美国冷泉港实验室、弗吉尼亚联邦大学、宾夕法尼亚州立大学从事博士后研究。主要研究早期胚胎发育、细胞命运决定与转变、癌症发生发展过程中的表观遗传调控机制。在Nature、Cell Stem Cell、Cell Research、PNAS、Genome Biology、Dev Cell、Adv Sci等期刊发表SCI论文110余篇。主持国家重大科学研究计划项目、国家自然科学基金重大研究计划项目与面上项目10余项。先后获得教育部“新世纪优秀人才”、上海市“浦江人才”、“曙光人才”、“东方学者”特聘教授项目资助。

研究领域:
基因在时空上的精确表达受各种表观遗传因素的调控。基因表达差异导致不同的表型,如高矮胖瘦、健康与生病。目前研究较多的表观遗传因素有DNA甲基化、组蛋白修饰、核小体定位、染色质高级结构、非编码RNA、RNA修饰与编辑等。我们课题组聚焦前四种表观四会因素在早期胚胎发育、细胞命运决定、重大疾病中的表观遗传调控机制。
一、早期胚胎发育中的表观遗传调控机制
精卵结合经过卵裂、增殖、分化,形成形态各一功能迥异的不同细胞。这些细胞组合形成组织,多种组织再进一步形成器官,最后形成一个个体。透过现象看本质,这些形态各一功能迥异的细胞都来自一个受精卵,拥有相同的一套基因。但在胚胎发育过程中,相同的一套基因受到精确的表观遗传调控,在时空上表达不同,从而形成形态各一功能迥异的不同组织细胞。在早期胚胎发育中,最初由卵子中带来的RNA与蛋白支撑受精卵发育,很快过渡到由合子基因组启动自身转录表达来支撑受精卵后续的发育,这一过程叫合子基因组启动(也叫母源到合子转变),它对早期胚胎发育至关重要,也受到精确的表观遗传调控。此外,在整个胚胎发育过程中,表观遗传基因调控出现异常,重则导致胚胎发育停滞,轻则导致出生缺陷,给社会与个人家庭带来极大负担与痛苦。我们通过正常受精与体细胞核移植的小鼠早期发育各阶段胚胎,利用多组学技术,通过生物信息学分析与实验验证,研究早期胚胎发育过程中的表观修饰图谱变化,鉴定关键蛋白因子,并探索其对早期胚胎发育的表观遗传调控机制。研究成果对理解和预防发育异常相关疾病有重大意义,在辅助生殖、畜牧业中优良品种及濒危物种的保种育种有重要应用价值。
二、细胞命运决定与转变中的表观遗传调控机制
胚胎干细胞具有全能性,能分化成生物体的各种细胞。反之,经过体细胞重编程,可把已分化的体细胞诱导成具有全/多能性干细胞。另外,对少数类型细胞,通过转分化技术,可直接把一种类型体细胞转变成另一种类型体细胞,即细胞转分化。在上述细胞命运决定与转变过程中,表观遗传调控发挥重要作用。我们课题组利用成熟的体外细胞分化平台、体细胞重编程技术与转分化技术,用二代测序技术测定不同阶段的基因表达、各种表观修饰、关键转录因子全基因组结合位点,通过生物信息学分析与实验验证,探索细胞命运决定与转变中的表观遗传调控机制。研究成果为高效获得高质量的干细胞、前体细胞、特定分化细胞提供理论依据与技术支撑,促进干细胞在基础研究与临床治疗中的应用。
三、癌症发生、发展、转移与耐药的表观遗传调控作用及机制
大量研究表明,许多重大疾病如癌症、老年痴呆等神经退行性疾病都与表观遗传基因调控紊乱有密切关系。癌症是最难治疗的疾病之一,严重危害人类健康。癌症之所以难治疗,是其具有转移与耐药的特性。二者皆与表观遗传调控有密切关系。我们课题组通过体外细胞水平、小鼠体内、与临床样本三个层次,探究癌症干细胞干性维持、癌细胞转移与耐药的表观遗传调控机制,鉴定关键蛋白因子与基因组位点,作为潜在的治疗与药物靶点。研究成果可改善我们对癌症发生、发展、转移与耐药调控机制的理解,最终促进临床上对癌症的诊断与治疗。

主持参与了国家“973”重大科学研究计划项目、国家自然基金委重大研究计划与面上项目。
1.“973”重大科学研究计划项目“胚胎发育中的核小体重排与染色质重塑”。
科研成果:
1. 鉴定、测序和注释了200多个Myb基因,并存入NCBI数据库作为共享资源。
2. 收集整合了哺乳动物中(人、小鼠、大鼠)36个与癌症关联的转录因子家族的成员蛋白因子,并鉴定到这些蛋白因子的共11660个目标基因,识别和标注出这些基因的启动子,以此为基础,构建与癌症关联的基因调控网络,成为癌症研究中的重要资源。
3. 通过单核苷酸多态性分析,证实哺乳动物CpG岛的丢失是由于新的DNA甲基化从岛的边缘开始,慢慢向岛中央“腐蚀”造成的。这一CpG岛丢失机制的证实将极大促进我们对癌症中DNA甲基化这一表观遗传学现象的了解,为设计和研发脱甲基化药物重新激活被沉默的基因作为治疗与DNA甲基化关联的癌症的一个手段打下基础。
4. 近年来利用染色质免疫沉淀结合高通量测序技术,首次在酵母和果蝇中绘制出高分辨率全基因组核小体图谱,并分析核小体定位对基因转录的调控机制等。设计开发了预测、分析核小体及其它蛋白的DNA结合ChIP-seq数据的软件GeneTrack。
5. 江赐忠教授的研究结果发表在Nature、Nat Rev Genet、Genome Research等国际著名学术刊物上,共发表20篇学术论文,并撰写专业书的一个章节。以第一作者发表的论文总影响因子超过103,以非第一作者发表的论文总影响因子超过59。其中,果蝇核小体图谱绘制与基因调控方面的研究成果以Article形式于2008年发表在Nature上,目前单篇引用已超过252次。同年在Genome Research上发表的关于酵母核小体与基因调控方面的研究报道,已被引用211次。2009年在Nature Reviews Genetics上发表了一篇关于核小体定位和基因调控的综述文章(作为该期封面文章),已被引用223次。2009年10月在Genome Biology上发表的酵母核小体系统参考图被评为“highly accessed”的文章。值得一提的是,发表在Nature和Genome Research上的两篇文章同时被国际著名学术期刊Nature Methods以“Research Highlights”形式进行报道;发表在Nature Reviews Genetics上的文章被国际著名学术期刊Nature Reviews Genetics以“Research Highlights”形式进行报道。
