马辉,现任清华大学深圳研究生院生命与健康学部教授、博士生导师。
教育及工作经历:
1982年吉林大学物理系,获学士学位。
1982-1983年长春光机所光学专业,获硕士学位。
1988年伦敦大学帝国理工学院物理系博士,专业方向为激光光谱、原子分子物理。
1988-1989英国伦敦大学帝国理工学院激光联合体博士后。
1989-1991年大连化学物理研究所分子光谱实验室博士后。
1991以后加入清华大学物理系原子分子纳米科学教育部重点实验室,任讲师、1993年晋升副教授、2001年晋升教授、博士生导师。
2003年4月加入清华大学深圳研究生院创建“光学检测与成像实验室”。
2003.3.20-2014.1.20清华大学深圳研究院副院长。
2014.1.20至今,清华大学深圳研究院生命与健康学部主任。

生物光子学(2006年之前):包括:共焦与双光子荧光成像、光学二次谐波成像、荧光各向异性度成像、荧光涨落谱,上述方法在生命科学研究中的应用。
生物医学光学(现在):基于光散射的无损成像方法及其应用研究,包括:
光学相干断层成像
偏振光散射理论、方法和应用,包括:偏振光散射理论、模型与模拟方法,偏振特征表征与提取方法,偏振敏感测量与成像方法,偏振光散射的应用。
研究方向:
1991-1997年以前从事专业方向为激光光谱技术和原子分子物理,1998年以后从事生物物理实验方法研究,研究方向为显微成像与微区荧光分析。2004年赴清华大学深圳研究生院创建“光学检测与成像实验室”,展开多项研究内容,包括:生物组织中偏振散射与传播行为研究、偏振光成像方法与应用研究、非线性光学显微成像方法与应用研究。
偏振光散射与应用研究:
偏振是光的基本特征之一,但也是多年来一直被忽视的一个特征。我们周边无处不在的光或多或少都有一些偏振成分,所有的物体都会改变光的偏振。偏振光的状态采用四分量斯托克斯(Stokes)向量描述,其中一个分量是光强。探测光的偏振态,可以用一个4x4穆勒(Mueller)矩阵描述所有物体的偏振光学性质。由于人眼对偏振不敏感,我们只能看到周边世界光强变化,即穆勒矩阵16个阵元中的一个。因此,偏振光测量包含更加丰富的信息。并且偏振信息对亚波长微观结构更加敏感。研究、发展偏振光散射测量方法,可以大大丰富我们对周边世界的感知能力。

研究内容
偏振光散射理论与计算模拟:利用各种单次散射理论,研究不同散射模型下偏振光在复杂介质中散射传播过程,特别是偏振指标与微观结构的对应关系。研究内容包括:单次散射理论与方法,散射模型,散射仿体,CPU与GPU模拟方法。
偏振光测量:研究不同偏振量测量方法和误差传递规律,包括:分时与同时穆勒矩阵测量方法与技术,偏振测量误差传递,对特异性结构敏感偏振量的测量方法。
偏振信息提取与特异性指标:获得特异性微观结构对应的偏振指标,包括:穆勒矩阵分解与变换,偏振态显示,大数据关联与统计分析。
偏振光散射应用:研究偏振散射方法在固体微观结构(生物组织、材料)、气体颗粒物和海洋微生物测量中的应用。

光学检测与成像实验室
实验室简介:
光学检测与成像实验室于2004年5月成立,2010被评选为深圳市重点实验室。实验室的科研队伍由四名教授、一名副教授、两名讲师、一名实验员和三十多名博士、硕士研究生组成。实验室在研究生院支持下,现拥有实验仪器设备固定资产超过800 万,包括荧光显微镜、多种半导体光源(包括宽带高功率SLD 和不同波长的多个全固化连续激光器)和超快激光光源、光电倍增管和制冷CCD等探测器、数字示波器,以及各类光学、光机械元器件和光学检测所需的一些基本科研装备。验实室面积近300 平方米,拥有光学平台9个、以及光学暗室、光电子学实验室、光学镀膜实验室以及机械加工等辅助设施,各类微机三十多台。
招生信息:
目前实验室每年招收理学硕士研究生5-7名;生物医学工程硕士研究生5-7名;理学博士研究生3-5名;除此之外,还招收清华大学-伯克利学院博士研究生2名,硕士生2-5名。

主要成果
2004年实验室成立以来已在国内外重要期刊上发表论文一百八十余篇,申请国家发明专利16项,美国专利1项。研制出适用于材料生产的在线监测、人工组织工程的在线监测、中医形态学诊断等领域的光学检测成像系统共6套,应用于临床眼科诊断成像系统已经转移到深圳市企业,产品已经推向市场。实验室成员获得2004年和2007年国家发明二等奖,2005年科学技术一等奖,2009年广西科技进步二等奖一项。

马辉:多个角度看世界
近年来,3D电影越来越受到人们的追捧。走进3D电影院,带上一副偏振眼镜,立体影像便展现在眼前。
其实,3D电影只是偏振应用的冰山一角。很少有人意识到,我们周边实际上是一个偏振光的世界。通过对偏振光性质的测量,我们可以检测区分不同的癌变组织,可以分辨不同粒径和成分的气体污染颗粒物,可以对海洋中的微生物进行现场计数与细致分类……而这些都是清华大学深圳研究生院教授马辉正在研究的内容。
光学方法是使用时间最长、基础积淀最深厚、应用范围最广泛,并且仍然迅速发展的观测手段。“照圣经的说法,上帝创造世界做的第一件事就是创造光。”马辉告诉记者,人类最早就是通过眼睛和光认识周边世界,随后用望远镜认识星空宇宙,用显微镜认识微观世界,光学测量方法的发展伴随着自然科学发展的整个历程。
如今,光学仍然是一个非常具有活力的科学领域,并不断获得诺贝尔奖。例如,2008年的绿色荧光蛋白(GFP)、2009年的光纤和CCD以及去年的LED和超分辨成像。“如果你把这些成果串起来,你会发现在这短短几年中,光学成像方法全链条各个环节都获得了诺贝尔奖:GFP对样品实现分子特异性对比度增强,LED是新型光源,光纤代表一种新型光路器件,CCD是新型探测器,而超分辨成像则是新的光学成像机理。”可以看出,古老的光学观测方法正不断焕发出新的生命力!2013年,联合国确定2015年为“光之年”。
偏振是光的一个属性,但很不幸,人类以及绝大多数动物的眼睛对光的偏振并不敏感。因此,尽管偏振这个词大家都耳熟能详,但偏振方法的潜力一直没有得到真正重视,做偏振研究的人很少,偏振的应用也很少。从2004年开始,马辉将偏振光散射这个看起来并不时髦的领域作为自己的研究方向。
十几年来,人们在越来越多的领域开始意识到偏振测量的潜力,马辉满怀自信地说:“偏振可以大大增加光学方法获取信息的能力,它将是光学发展的新方向,在信息技术突飞猛进的今天,偏振方法前景无限!”。
实现癌症早期检测
马辉告诉记者,光学方法很适合于生物样品。它基本不产生损伤,分辨率可以达到亚细胞层次,还可用于从活细胞到组织、器官和个体等不同层次的生物样品。“但生物组织是不透明的,因为它有散射,当激光照射进人体,光能透进去,但看不清里面的血管和骨头。散射破坏了光的有序性质,这是光学方法用于生物组织最大的问题。”马辉解释道,“利用偏振方法,可以抑制多次散射退偏光子对测量结果的贡献,提高生物组织浅表层测量的分辨率。”
马辉最开始研究偏振方法的一个基本目的就是降低散射的影响,提高成像质量。“人们发现,85%以上的癌变首先在上皮组织中产生,提高表皮组织成像的分辨率有可能帮助医生发现早期癌变。”
偏振测量的另一个功能是获得生物组织微观结构的信息。马辉告诉记者,散射可以改变光的方向,破坏成像结果,但散射也可以影响光的偏振,带来新的信息。通过研究散射光偏振的变化与散射体的大小、密度、形状和结构之间的关系,人们可以获得样品丰富的微观结构信息,由此发现生物组织的异常和病变,实现病理诊断。
此外,偏振可以帮助提高浅表层组织成像的分辨率,还能够测量生物组织微观结构的变化,在癌症早期诊断方面具有很好的应用前景。马辉介绍,他们正与深圳南山医院病理科合作测量癌变组织。“全偏振成像叫穆勒矩阵成像,我们从医院找来很多不同类型的癌变样品做穆勒矩阵成像,包括皮肤基底细胞癌、甲状腺乳头状腺癌、乳腺癌以及宫颈癌等等。现在我们已经有一些证据,能证明全偏振成像的确有可能用于癌变的定量表征,但是我们还需要继续搜集大量的样本积累数据。”
2015年,马辉获得了国家重大科研仪器研制项目,他将利用这几年的最新基础研究成果和技术进展研发模块化穆勒矩阵显微镜,演示其典型临床应用。同时,他还将建立实验室研究平台,针对各个模块开展原理和方法学探索和工程开发,研究信息提取方法和大数据分析方法,不断改进仪器,推动产业化。
“2014年,超分辨成像的其中一个诺贝尔奖获得者赫尔,首先在激光成像中实现了超分辨,但他采用的方法很复杂,不好使用。但赫尔很机灵,他让莱卡公司把他的方法做成了仪器,给生物学家用。尽管这个仪器现在仍然要卖一千万人民币,并且仍然不那么好用,但由于生物学家展示了他的巨大应用价值,白兹格利用贝尔纳的工作成果实现了另一种更好用、更便宜的光学超分辨成像方法,最后三个人一起得奖。”马辉总结道,“光有好概念、好方法还不行,必须推广出去,让企业介入把方法变成产品,供更多的人使用。我这个仪器项目就是要发展一系列方法和装置,并且拉上企业做仪器,为生物学家和医生提供新的工具,提出新的需求,带动偏振方法的研究。”
作为探索者,马辉一路走来碰壁不少,他说:“走很多弯路之后最后发现碰对了,也是一种享受。我觉得做科研就要经历这些过程!我会沿着这条路继续往下走,把偏振的潜力不断挖掘出来。”
偏振开启一扇门
马辉做偏振成像和应用是半路出家,他1988年在英国帝国理工学院获得博士学位,1989年回国,后来加入清华物理系工作,有十几年时间他一直从事原子分子光谱基础研究。1995年对马辉来说是一个转折点。这一年,他在美国麻省理工学院访问,与世界顶级科学家合作,研究双原子分子高激发态光谱。
这一年中,马辉参观了很多实验室。“我在麻省理工学院了解到利用拉曼光谱发现癌变和利用光学相干断层成像测眼底的工作,在麻省综合医院参观了交叉学科的威尔曼实验室,在阿贡国家实验室参观了高分子光电转换材料研究组,这些都给了我非常深刻的印象。”这些交叉学科前沿研究让基础研究出身的马辉思索起来,“那一年我确实开始有些想法,觉得自己还是喜欢做偏应用一些的研究”。
回国后马辉逐步把工作重点转向生物物理实验方法研究。从1998年到2003年,他率先在国内开展双光子荧光成像和光学二次谐波成像,与国际上基本同步。他还建立了荧光涨落谱分析方法,并在荧光偏振成像方面开始探索,“我觉得那时候还真是进展很快,每天都有新的东西”。
2003年,马辉被调到刚刚建立的清华大学深圳研究生院。深圳是中国改革开放的窗口,有突出的创新文化和快速成长的高技术产业,但高等教育却滞后于经济发展。“深圳有着一个很强、很完善的产业链结构,而且非常重视应用。在深圳做事,不得不考虑创新将来可能用在哪,能产生多大的影响。”脱离于北京装备精良的实验室,马辉重新思索科研方向。“深圳研究生院有两个特点,一个是产业合作,一个是学科交叉。既然来到深圳,就必须充分利用这些特点。”
2004年,从英国回来的一位老师加入了马辉课题组。“他曾做过OCT方面的研究,于是我们决定做OCT产业化。为了同OCT的研究互相呼应,我也开始进行偏振光散射及其应用研究,这是一个全新的研究方向!”
马辉向记者介绍,OCT就是光学相干断层扫描仪的简称,它可以通过背向散射对眼底视网膜或眼角膜进行微米量级高分辨成像,为医生提临床诊断信息。课题组与当地公司合作,开启了OCT产业化之路。
时至今日,当初合作的小公司已经占据中国眼科OCT产品30%以上市场份额,实验室毕业的几个硕士也成为了公司的骨干。与此同时,针对偏振光散射的研究也日益深入,从最初一个应用小课题,逐步产生新问题,带出新模型、计算程序、实验方法和应用,形成了完整的研究方向。回想整个发展历程,马辉十分感慨:“当初做偏振成像的时候我并没有很高的期待值,周围朋友也不认为它有多大的前途,但深入做下去就闯出一条路,而且可能是一条康庄大道。我们周围是一个偏振光的世界,当你能感受偏振的时候,你会发现这个世界如此丰富多彩!”
最近,偏振研究越来越引起人们的关注,马辉带领团队坚持了10余年,已经形成自己的研究特色,在国际上崭露头角。目前马辉在偏振上专注于做四件事,“第一是测量,我们发展了一系列方法,能够测得更加准确和迅速;第二是信息提取,就是针对不同应用寻找一些具有特异性的偏振指标,比如某种癌变的临床诊断指标;第三是信息解释,就是要知道不同微观结构如何影响散射光偏振态变化,为解决逆问题提供支撑;第四就是应用,千方百计把偏振用于解决各种实际问题。应用也是我认为最重要的一件事”。
做一个牵线人
对马辉来说,偏振能为我们提供一个信息宝藏。他打了一个形象的比喻,“不考虑偏振的时候,我们相当于用一个字母来给不同人命名,也就是只能用强度编码。甲叫a,乙叫aa,丙叫aaa……而偏振能让我们用单词命名,jia、yi、bing……穆勒矩阵的16个阵元相当于给了我们16个字母,可以形成各种组合,我们描绘周边世界的能力一下子提高很多”。另外,偏振方法具有普适性,现有光学方法都可能通过偏振调制升级为偏振方法,应用于不同类型的问题和样品。
2013年,“雾霾”成为年度关键词,世界上污染最严重的10个城市,有7个在中国。为此,我国在雾霾高发的16个省份选择了43个监测点,以期掌握不同地区PM2.5污染特征及成分差异。“中国的污染很复杂,往往不知道污染源是什么,我们正在同一家公司和环保部门合作,发展一种利用偏振散射对空气中颗粒物进行计数、分类和源解析的仪器”,他进一步解释,“这就像一个农贸市场,原先我们只能估算人流总量,现在能告诉你不同时刻男人、女人、小孩各有多少,便于优化管理。”
几年前,马辉参与了国家海底观测试验网项目,负责数据管理系统。专业敏感性使马辉开始思考如何将偏振应用到海洋,“通过这个项目我知道了很多海洋方面的需求,我们可以利用偏振方法升级现有的光学技术,提高通过测量所能获得的信息量”。
从上世纪80、90年代开始,全球平均气温比100年前上升了0.48℃,科学家们针对全球变暖这一自然现象提出了种种猜测。马辉告诉记者,“其实藻类对环境变化影响很大,通过监测海洋里不同类型的藻类,将不同海域里不同藻类的浓度等测出来,科学家就能探究气候变暖的原因”。
马辉常将自己的工作比喻为“造枪”,“我们就是要提供新型手段,帮助别人获得信息。我就像一个造枪的,我不打仗,但我帮助打仗的人,提升军队的战斗力”。
从事科学研究,兴趣莫过于最好的助推剂。“我觉得发现问题之后解决问题很好玩,我就是喜欢做科研。”虽然时常奔波于全国各地甚至国外,但马辉说:“这些年为科研而奔波,我还是挺享受的。”
在马辉眼中,偏振好似一颗正在发芽的种子。数年来,在他的悉心呵护下,这颗种子破土而出,渐渐长大。他相信偏振在未来的应用潜力终将被更多人认知,就像彩色电视替代了黑白电视,偏振下的新景色将给人类带来一个翻天覆地的变化。
